“金桃”猕猴桃胚乳植株再生体系的优化_胚乳培养-论文网

4结论

在猕猴桃“金桃”品种的胚乳愈伤组织诱导过程中,2,4-D的诱导作用最显著,培养基1/2MS+1mg/L2,4-D+3mg/LBA+20g/L蔗糖是最佳的诱导培养基。从愈伤组织分化不定芽,需要一个不含2,4-D的培养过程。而在诱导不定芽分化过程中,高浓度的NAA对不定芽分化有一定抑制作用,当BA浓度为5μmol/L时,最适合不定芽的分化。同时低浓度的蔗糖(20g/L)和适当时间的暗培养(7d)能显著地促进不定芽的形成。应用流式细胞仪鉴定了胚乳愈伤组织诱导得到的植株染色体倍性。
