此外,CK处理的细胞内比较干净,在细胞壁、细胞膜、线粒体等系统内没有发现多余物质存在(图1、图2、图4、图5),但3种钙处理中,少量细胞内较干净(图14),但大量细胞内有较多的黑色物质存在(图7、图10),不均匀的分布于细胞壁、细胞膜、液泡膜、线粒体等系统中(图8、图9、图11、图12、图13、图15、图16)。这些黑色物质的成分并不明确,可能为一些多酚类的物质。此外,3种钙处理之间的细胞超微结构差异并明显。
3结论与讨论
3.1樱桃对钙的吸收
本文测定了樱桃果肉中的全钙含量及不同提取剂(80%乙醇、蒸馏水、1mol·LNaCl、2%HAC和0.6mol·LHCl)逐级提取的不同形态钙含量,可以看出总钙含量及不同形态钙的含量差异。在果肉中钙的各种形态中,ALc-Ca是主要的存在形式,其含量占全钙的34.7%~47.2%,而ALc-Ca主要以硝酸钙、氯化钙以及氨基酸钙的形态存在,由此我们可以断定樱桃果肉中主要以这几种形式中钙的一种或者几种形态存在,至于具体是何种具体的形态,虽然本试验研究结果中并没有涉及,但在3种不同的钙处理中,Che-Ca处理中的全钙含量最高,ALc-Ca所占的比例也最大,而Che-Ca中的钙主要以螯合态存在,根据这一点我们可以推断樱桃吸收的钙中,螯合态钙是相对比较容易吸收利用的形态,而在果肉中钙的各种形态中,氨基酸钙应当也占有相当的比例。
3.2樱桃贮藏后的钙的转化
本试验结果显示,樱桃贮存一段时间后果肉中总钙含量也有小幅下降,有人研究苹果果实在贮存期间果肉中的钙会向果皮转移,而樱桃可能也存在着类似的现象。贮藏前后各种形态钙含量发生了不同规律的变化,这说明果肉细胞内钙库与细胞外钙库在贮藏期间处于动态平衡状态。樱桃贮藏60d后,HAC-Ca、HCl-Ca的含量有所上升,ALc-Ca、NaCl-Ca和HO-Ca均有不同程度的下降,也就是说易溶的离子态钙和果胶酸钙含量下降,磷酸钙、草酸钙等难溶形态的钙含量有所增加。苹果在贮藏期的软化与钙溶性的转化有关,易软化的苹果水溶性钙含量明显降低,董彩霞分析认为果胶酸钙等易溶态钙是细胞壁和细胞膜稳定的重要因素,其含量降低必将导致细胞壁和细胞膜系统的不稳定。樱桃贮存60d后,产生的一些有机酸类物质导致了樱桃的可滴定酸度降低,这些有机酸类物质同各种易溶态离子钙相结合,形成了草酸钙、磷酸钙等难溶态钙,这也许是导致NaCl-Ca、HO-Ca和ALc-Ca含量下降,而HAC-Ca、HCl-Ca的含量上升的原因之一。
3.3贮藏后超微结构
在果实成熟衰老过程中,常伴随着细胞结构的变化。本实验樱桃在贮藏60d后细胞超微结构的电镜扫描结果中,CK处理膜系统受到不同程度的损坏,并且部分细胞有了收缩的现象。而3种钙处理的细胞结构比较完整,但有部分黑色物质不均匀的分布于细胞壁、线粒体、液泡膜等系统中,尤其是Che-Ca处理。这些黑色物质可能为一些多酚类的物质,具有抗氧化、抗衰老等作用,延缓了樱桃贮存期间病害的发生。有资料在苹果中证实Ca是一种膜保护剂,膜结构损伤,能导致液泡中的钙流入细胞质;膜功能损伤,引起运输Ca和Ca-ATP酶和Ca/nH反向传递系统功能受到破坏,进入胞质的Ca难以流出,而本试验的电镜扫描照片也显示,钙处理对樱桃贮藏期间膜系统也有一定的保护作用,对破坏膜结构和功能的因素有一定的拮抗作用。充足的水溶性及果胶酸钙可保证了果实的正常生理功能,从而可有效的延缓果实贮藏期间的软化及病害的发生。而3种不同形态钙之间造成的差异可能与3种钙制剂的分子结构及樱桃对钙的需求特点有关,其具体原因还有待于做深一步的研究。以上分析我们知道樱桃在贮存过程中有效钙是不断减少的,因此可以认为钙的无效化和由此引起的细胞膜系统的结构和功能的损伤是樱桃贮存期间质量下降的重要机理之一。
